Il destino ha riunito un eterogeneo gruppo di viaggiatori sul lussuoso battello da crociera Karnak, in navigazione sul Nilo. La personalità dominante è senz'altro Linnet Ridgeway, la ragazza più ricca d'Inghilterra, dotata di grande fascino e abituata a essere sempre al centro dell'attenzione. Attorno a lei gravitano un fidanzato respinto e diversi accaniti ammiratori in competizione tra loro. Ciascuno dei personaggi ha però una sua storia e un suo segreto da custodire, ben nascosto sotto la facciata di rispettabilità e di perbenismo. Fra i turisti c'è anche Hercule Poirot, una volta tanto in vacanza, ma anche in questa occasione il suo ozio è destinato a durare poco. Nel giro di poche ore, infatti, a bordo del Karnak si consumano ben due delitti, e la tranquilla crociera si trasforma in una disperata caccia all'assassino.
