Nei cinque libri della sua opera - adesso pubblicati con il testo francese in edizione critica a fronte - il suo pubblico ha visto di volta in volta il puro intento comico e burlevole, l'atteggiamento irreligioso e dissacrante, la volontà di richiamarsi a una cultura popolare e carnevalesca, il recupero dei valori materiali e corporei, l'adesione a un cristianesimo vicino all'evangelismo erasmiano. Tutte queste letture hanno una parte di verità. Ma soprattutto va tenuto presente l'esplicito intento di Rabelais di proporre quello che egli stesso chiama il "sostanzioso midollo" dell'opera: una trama simbolica che, sotto la superficie comica, coincide coi percorsi del dubbio e si risolve nella ricerca della verità e della saggezza.
